Non-invasive detection of universal osteodifferentiation signatures through untargeted NMR Exometabolomics

abstract

The secretion profile of mesenchymal stem cells (MSCs) is a major driver of their therapeutic effects, although secretome metabolites are largely unexplored. Exometabolomics may reveal new information on MSCs, providing means to monitor osteodifferentiation. Here, untargeted NMR metabolomics was applied to monitor exometabolome of human adipose-derived MSCs from 3 different donors, throughout 21 days of osteodifferentiation. Results revealed changes in metabolic activity, including energy metabolic switch, cell membrane metabolism and antioxidative protection. The donor-independent features may be exploited to assess cell osteogenic ability and advance metabolic candidates for media optimization.
